Det er 1943, og et commando-team sendes på en selvmordsmission mod en tysk befæstning i det græske øhav. Teamet er forfulgt af uheld fra landgangen på lodrette klippesider i orkan til tyskernes præcise viden om deres planer og indbyrdes had mellem de amerikanske, engelske og græske soldater. Men kaptajn Malloy presser sine mænd mod det umulige mål: at ødelægge de to tyske kæmpekanoner, som ellers vil udslette en engelsk flåde-eskadre på vej mod strædet ud for Navarone.
