18-årige Ian (Josh Zuckerman) fra Chicago har ikke det store held med damerne. Derfor bliver han også temmelig overrasket, da en laber blondine på internettet tilbyder at tage hans mødom, hvis han kører fra Chicago til Knoxville og møder hende. Ian kaster sig ud i vognen sammen med sine to venner og jokker sømmet i bund. Men vejen er lang, og trioen bliver mødt af den ene mærkværdige forhindring efter den anden, bl.a. den bilkyndige Ezekiel (Seth Green), der er medlem af amish-samfundet.
