I 1500-tallet begiver Aguirre sig på jagt efter det mystiske El Dorado. Sammen med en spansk ekspedition drager han ud for at finde det sagnomspundne guldrige i Amazonas urskov. Den tyranniske Aguirre begår mytteri mod gruppens leder og overtager kommandoen i sin besatte søgen efter guld i rigelige mængder. Hans virkelighed bliver mere og mere sindssyg, og i en magtgal vision bestemmer han sig for at grundlægge et nyt kejserligt imperium.
