Året er 1909 og den tyske opdagelsesrejsende Theodor Koch-Grünberg har begivet sig ind i den colombianske jungle for at finde shamanen Karamakate, der er en af de sidste overlevende medlemmer af Ocainas-stammen. Theodor er overbevist om, at Karamakate kan hjælpe ham med at finde den hellige Yakruna-plante, hvis helende kræfter den alvorligt syge Theodor desperat har brug for. Karamakate er skeptisk over for Theodor, som han er over for alle hvide mænd, men indvilliger i at hjælpe ham, da han påstår, han kan lede Karamakate på sporet af andre tilbageværende stammefæller.
