Det er en udsøgt skare, der mødes på Orientekspressens perron i Istanbul. Man hilser, og man konverserer, men før toget når Paris, er den fornemme overflade krakeleret. En af passagererne dør en frygtelig død. Heldigvis er den skarpsindige belgiske detektiv Hercule Poirot også med toget, og han giver sig straks i kast med opklaringsarbejdet.
