Historien foregår på planeten Lythion i år 40.000 e.Kr, hvor Barbarella tvinges til at lande på sin rejse gennem rummet. Hun optræder som en kvindelig James Bond, der besejrer ondskaben i form af robotter og monstre. Hun belønner også hæmningsløst de flotte mænd, som hjælper hende i eventyret. Uanset om hun bryder med de sorte vagter, den onde dronning eller englen Pygar, syntes det naturgivet, at hun skal miste mindst én del af sin kropsnære rumdragt!
