The Knick, som skildrer den intense hverdag på The Knickerbocker Hospital i New York. I begyndelsen af det 19 århundrede tester dristige læger en lang række nye metoder, der skal redde liv. Odds’ene er tvivlsomme og medicinen har stadig mange mangler. Hospitalet står overfor en lang række problemer – de velstående patienter erstattes gradvist af fattige patienter og direktøren Herman Barrow håndterer ikke hospitalets økonomi ansvarsfuldt. Men så dukker den dygtige kirurg John Thackery op, og han har ingen moralske skrupler.
