Godine 1539. malteški su vitezovi namjeravali španjolskom kralju Karlu V. darovati statuu zlatnog sokola ukrašenog rijetkim draguljima. No gusari su presreli galiju te je 'Malteški sokol', čini se, nestao zauvijek... San Francisco, četiri stoljeća kasnije. U detektivsku agenciju Spade & Archer koju zajednički vode Sam Spade i Miles Archer dolazi gospođica Wonderly iz New Yorka tražeći da joj pomognu pronaći nestalu sestru. Iako obojica posumnjaju u njezinu priču, prihvate posao zadovoljni što njihova nova klijentica plaća gotovinom. Kad Archer uskoro bude ubijen, Spade shvati da se upleo u opasan posao pri čemu dozna da je gospođici Wonderly pravo ime Brigid O'Shaughnessey. Istraga ga dovede do dvojice prevaranata Joela Caira (P. Lorre) i Kaspera Gutmana (S. Greenstreet) koji kao i Brigid tragaju za 'Malteškim sokolom'.
