Nikita er en narkotikaavhengig ungdomskriminell som ble anklaget for å ha drept en politimann i kaldt blod under et forsøk på ran av et apotek. Hun blir senere arrestert og dømt til døden ved giftsprøyte, hvor hun i hemmelighet ble dopet av myndighetene, og iscenesatt sin død. Nikita blir deretter «rekruttert» av en hemmelig regjeringsorganisasjon og forvandlet til en høyt kvalifisert leiemorder som ikke kan spores.
